Significance of Receptor Mobility in Multivalent Binding on Lipid Membranes

چکیده

Numerous key biological processes rely on the concept of multivalency, where ligands achieve stable binding only upon engaging multiple receptors. These processes, like viral entry or immune synapse formation, occur diffusive cellular membrane. One crucial, yet underexplored aspect multivalent is mobility coupled Here, we discuss consequences in from four perspectives: (I) The facilitation receptor recruitment by ligand due to their diffusivity prior binding. (II) effects preassembly, which allows local accumulation. (III) changes formation receptor/ligand complex. (IV) lipid environment surrounding engaged We demonstrate how understanding essential for fully unravelling principles membrane leading further development studies interactions, and guide design new generations ligands.

Lipid mobility and molecular binding in fluid lipid membranes.

The lateral mobility of dilute concentrations of fluorescently labeled lipids doped into supported membranes is found to change upon receptor ligand binding at the membrane surface, even when the lipid is not directly involved in the binding event. Experiments using membrane microarrays are performed that illustrate the use of lipid mobility measurements as an effectively label-free strategy of...

Multivalent ligand-receptor binding on supported lipid bilayers.

Fluid supported lipid bilayers provide an excellent platform for studying multivalent protein-ligand interactions because the two-dimensional fluidity of the membrane allows for lateral rearrangement of ligands in order to optimize binding. Our laboratory has combined supported lipid bilayer-coated microfluidic platforms with total internal reflection fluorescence microscopy (TIRFM) to obtain e...

Binding of prion proteins to lipid membranes.

A key molecular event in prion diseases is the conversion of the normal cellular form of the prion protein (PrPC) to an aberrant form known as the scrapie isoform, PrPSc. Under normal physiological conditions PrPC is attached to the outer leaflet of the plasma membrane via a GPI-anchor. It has been proposed that a direct interaction between PrP and lipid membranes could be involved in the conve...

Binding of serotonin to lipid membranes.

Serotonin (5-hydroxytryptamine, 5-HT) is a prevalent neurotransmitter throughout the animal kingdom. It exerts its effect through the specific binding to the serotonin receptor, but recent research has suggested that neural transmission may also be affected by its nonspecific interactions with the lipid matrix of the synaptic membrane. However, membrane-5-HT interactions remain controversial an...

Significance of microRNA targeted estrogen receptor in male fertility

Objective(s): Estrogen receptor-alpha (ERα) mediates estrogen action in regulation of different levels of the hypothalamic-pituitary-testis axis. It has a key role in spermatogenesis. Estrogen receptor alpha knock-out (ER koα) male mice were infertile and severe impairment in spermatogenesis and seminiferous tubules was observed. Recently, it has been reported that microRNA (miRNA) mir-100 and ...
